Ordering Notes
For crossmatch testing, an order for recipient HLA Antibody Testing is needed.

Specimen and Container/Tube
Specimen Type: Whole blood and serum
Preferred Specimen: ACD solution A (2) and gold top SST (2)
Other Acceptable Specimens: For serum only: Tiger top (marble) SST, red top
Volume
Preferred Volume: 20mL whole blood (ACD), 6mL (serum)
Minimum Volume: 20mL whole blood (ACD), 6mL (serum)
Specimen Stability
Whole blood specimens may be kept at room temperature for up to 72 hours following collection.
Serum specimens may be kept at room temperature for up to 24 hours following collection. Once centrifuged, specimens may be kept at refrigeraton (4°C) for up to one week.

Collect specimen per standard laboratory collection procedures.
Specimen Handling and Transport Instructions
UCM Onsite Instructions: Collect specimen per standard collection procedures and send to the laboratory immediately.
UCM Offsite and UC MedLab Outreach Instructions: Collect specimen per standard collection procedures and send to the laboratory using standard transport procedures.
Rejection Criteria
Whole blood specimens greater than 72 hours old may be rejected; cellular viability may not be adequate for testing.
Unspun serum samples greater than 24 hours may be rejected. Sample with visible signs of hemolysis (H-Index > 100) may be rejected.
Reference Values
No reference range; results may be provided in an interpretive report.

Turnaround times are relative to the time the specimen is received in the testing laboratory section.

Clinical Indications
The flow cytometric crossmatch is a test that assesses the level of circulating antibodies in the recipient that are directed against the HLA class I and/or class II molecules of the recipient cells (self). This test may determine if a recipient’s autoantibodies are responsible for a crossmatch result, and can be used to troubleshoot unexpected reactivity from other methods.
